以1次、半巢式及多重PCR方法扩增58例淋巴细胞白血病IgH及TCRVγI-Jγ克隆性基因重排。结果显示:多重PCR和1次PCR的敏感度为10 ̄(-4)~10 ̄(-5)水平;半巢式PCR敏感度可达10 ̄(-5)~10 ̄(-6)水平。IgH和TCRVγI-Jγ重排阳性率为67.2%和62.0%;半巢式PCR检测IgH重排阳性率为70.7%。上述结果表明:多重PCR可同时检测两对目的基因,适于初治病例检测,而对缓解病例的检测则需采用敏感度较高的半巢式PCR方法。
Fifty-eight cases of lymphocytic leukemia IgH and TCRVγI-Jγ clonal gene rearrangements were amplified by 1-step, semi-nested and multiplex PCR. The results showed that the sensitivity of multiplex PCR and primary PCR was 10 ~ (-4) ~ 10 ~ (-5), and the sensitivity of semi - nested PCR was about 10 ~ (-5) ~ 10 ~ (-6) . The positive rates of IgH and TCRVγI-Jγ rearrangements were 67.2% and 62.0%, respectively. The positive rate of IgH rearrangement detected by semi-nested PCR was 70.7%. The above results show that multiplex PCR can detect two pairs of target genes at the same time and is suitable for the detection of naive cases, while the detection of remission cases requires the use of highly sensitive semi-nested PCR.
