目的 研究氢化可的松对体外培养的大鼠睾丸间质细胞 (Leydigcells)睾酮分泌的影响 ,探讨氢化可的松影响睾酮分泌的作用机制。方法 将雄性大鼠处死 ,取睾丸间质细胞悬液分别加入氢化可的松、促性腺激素、雄烯二酮进行体外培养 ,用放射免疫分析方法测定睾酮含量。结果 氢化可的松可抑制促性腺激素刺激的睾酮分泌 ,但对睾酮的基础分泌及雄烯二酮升高的睾酮水平无影响。结论 在睾丸间质细胞 ,氢化可的松以剂量依赖性方式抑制促性腺激素刺激的睾酮的合成 ,但对细胞中 17β-羟甾脱氢酶的活性无影响
Objective To study the effect of hydrocortisone on the testosterone secretion in cultured rat Leydig cells and to explore the mechanism of hydrocortisone on testosterone secretion. Methods Male rats were sacrificed and the testicular stromal cell suspension was added with hydrocortisone, gonadotropin and androstenedione for in vitro culture. The content of testosterone was determined by radioimmunoassay. Results Hydrocortisone inhibited gonadotropin-stimulated testosterone secretion, but had no effect on basal secretion of testosterone and increased testosterone levels of androstenedione. Conclusions In testicular stromal cells, hydrocortisone inhibits gonadotropin-stimulated testosterone synthesis in a dose-dependent manner but has no effect on the activity of 17β-hydroxysteroid dehydrogenase
