目的 :探讨厚朴酚联合5-氟尿嘧啶(5-fluorouracil,5-FU)对人结肠癌HCT-8细胞增殖和分泌型卷曲蛋白4(secreted frizzled-related protein-4,SFRP-4)表达的影响。方法 :分别采用厚朴酚、5-FU和厚朴酚联合5-F U处理结肠癌HCT-8细胞后,MTT法检测细胞的增殖情况,Hoechst 33258染色法检测细胞凋亡的形态变化,FCM法检测细胞的周期分布,蛋白质印迹法检测细胞中SFRP-4和β-catenin蛋白的表达水平。结果 :厚朴酚、5-F U和厚朴酚联合5-FU均可抑制HCT-8细胞的增殖(P<0.05),其中联合用药组的抑制作用更强。厚朴酚、5-FU和厚朴酚联合5-FU作用后,HCT-8细胞出现明显的细胞凋亡的形态变化,联合用药组细胞凋亡的形态变化更明显。5-F U或厚朴酚联合5-F U作用于HCT-8细胞后,G0/G1期细胞所占比例增加,S期和G2/M期细胞所占比例下降(P均<0.05)。厚朴酚、5-FU和厚朴酚联合5-FU组HCT-8细胞中SFRP-4的表达水平明显高于未用药处理的空白对照组(P<0.05),5-FU联合厚朴酚组HCT-8细胞中β-catenin的表达水平显著低于5-FU组和厚朴酚组(P<0.05)。结论 :厚朴酚联合5-FU能显著抑制人结肠癌HCT-8细胞的增殖,增加G0/G1期细胞所占比例,其机制可能与调节HCT-8细胞中SFRP-4和β-catenin蛋白的表达水平有关。
Objective: To investigate the effects of magnolol combined with 5-fluorouracil on the proliferation and secretion of secreted frizzled-related protein-4 (SFRP-4) in human colon cancer HCT-8 cells . Methods: The proliferation of human colon cancer HCT-8 cells was treated with magnolol, 5-FU and honokiol combined with 5-FU respectively. The cell proliferation was detected by MTT assay. The morphological changes of apoptosis were detected by Hoechst 33258 staining. The cell cycle distribution was detected. The expression of SFRP-4 and β-catenin in the cells was detected by Western blotting. Results: Magnolol, 5-F U and honokiol combined with 5-FU all inhibited the proliferation of HCT-8 cells (P <0.05), and the combined treatment group showed stronger inhibitory effect. Magnolol, 5-FU and honokiol combined with 5-FU had obvious morphological changes of apoptosis in HCT-8 cells, and the morphological changes of apoptotic cells in combination group were more obvious. 5-F U or honokiol combined with 5-F U increased the proportion of cells in G0 / G1 phase and decreased the proportion of S phase and G2 / M phase in HCT-8 cells (all P <0.05). The level of SFRP-4 in the cells of honokiol, 5-FU and honokiol combined with 5-FU group was significantly higher than that of the untreated control group (P <0.05) The expression of β-catenin in HCT-8 cells was significantly lower than that in 5-FU and honokiol groups (P <0.05). CONCLUSION: Magnolol combined with 5-FU can significantly inhibit the proliferation of human colon cancer HCT-8 cells and increase the proportion of G0 / G1 phase cells. The mechanism may be related to the regulation of SFRP-4 and β-catenin proteins in HCT-8 cells The level of expression.
