目的观察动物感染广州管圆线虫后肺组织病理改变,以及应用抗广州管圆线虫成虫单克隆抗体进行感染鼠肺免疫组织化学研究。方法20只大鼠实验室人工感染广州管圆线虫,组织学方法观察肺组织的形态学变化,免疫组织化学方法应用实验室制备的抗广州管圆线虫成虫可溶性抗原单克隆抗体IgG和IgM进行肺组织切片的免疫反应性研究。结果受感染的大鼠肺组织肿胀实变,表面及切面质硬粗糙,有灰白色虫卵结节,病变范围广泛。HE染色镜下观察可见肺组织中有多个圆形、椭圆形的虫卵结节,结节周围组织细胞反应及纤维化,肺泡隔增厚,肺泡轮廓消失。结节内虫卵发育形成桑葚期、仔虫期及一期幼虫。用抗广州管圆线虫成虫单克隆抗体IgG和IgM分别行免疫组化,结果显示桑葚期虫卵表达IgG,而IgM则在桑葚期、仔虫期及一期幼虫等部位均有显著阳性表达。结论广州管圆线虫感染所致大鼠肺脏病理改变,主要表现在肺内形成多个虫卵结节,并可致肺呈纤维化改变;抗广州管圆线虫成虫单克隆抗体IgG和IgM在肺组织虫卵结节内的阳性表达有显著差别。
Objective To observe the pathological changes of lung tissue after the infection of Anemarr: angiostrongyliasis in Guangzhou and to study the immunohistochemistry of murine lung using the monoclonal antibody against adult Aphis gossypii. METHODS: Twenty-five rats were artificially infected with A. cantonensis. The histological changes of lung were observed by histological method. Immunohistochemistry was performed by using immunohistochemistry in laboratory. The monoclonal antibodies IgG and IgM of soluble antigen against C. elegans Tissue section immunoreactivity studies. Results The lungs of infected rats swollen and solidified, the surface and the section were hard and rough, with gray egg nodules and extensive lesions. Hematoxylin-eosin staining showed that there were many round and oval egg nodules in the lung tissue, the reaction and fibrosis of the tissues around the nodules, the alveolar septum thickening and the disappearance of the alveolar outline. Nodules within the formation of eggs develop mulberry stage, the stage and a larva. The results of immunohistochemistry with monoclonal antibodies IgG and IgM against the adults of A. cantonensis showed that the expression of IgG in the mulberry eggs was significant, while IgM was significantly expressed in the mulberry stage, the stage of the cord stage and the stage Ⅰ larva. Conclusions The pathological changes of the lungs in rats induced by C. elegans infection are mainly caused by the formation of multiple egg nodules in the lungs and fibrosis of the lungs. The monoclonal antibodies IgG and IgM against adults of A. elegans in the lung Tissue egg nodules within the positive expression were significantly different.
