目的:探讨胞内模式识别受体NOD2表达上调对于人早期妊娠蜕膜基质细胞基本生物学行为的影响。方法:分离培养人早期妊娠蜕膜基质细胞运用CCK8试剂盒、Transwell侵袭小室、流式细胞术等分别检测接受NOD2配体处理后细胞增殖、侵袭和凋亡能力。结果:蜕膜基质细胞接受相应配体刺激后NOD2 mRNA及蛋白表达水平上调,抑制细胞侵袭能力促进细胞凋亡,但是对细胞增殖能力无明显影响。结论:NOD2过度激活损伤蜕膜组织,可能导致不良妊娠结局。
Objective: To investigate the effect of upregulation of intracellular pattern recognition receptor NOD2 on basic biological behavior of human decidual stromal cells in early pregnancy. Methods: The decidual stromal cells of human early pregnancy were isolated and cultured. The cell proliferation, invasion and apoptosis were detected by CCK8 kit, Transwell invasion chamber and flow cytometry. Results: The decidual stromal cells were upregulated NOD2 mRNA and protein expression by the corresponding ligand stimulation, inhibit cell invasion ability to promote apoptosis, but had no significant effect on cell proliferation. Conclusion: Overexpression of NOD2 can damage the decidual tissue and may lead to adverse pregnancy outcomes.
