目的研究灵芝多糖对模拟AD学习记忆障碍大鼠脑组织白细胞介素-6表达(IL-6)的影响。方法采用双侧海马内一次性注射β-淀粉样多肽25~35片段(Aβ25~35)制作模拟AD学习记忆障碍大鼠模型,造模24h后治疗组腹腔注射灵芝多糖注射液,其余各组分别注射等量的生理盐水,1次/d,疗程7d,并于第8d开始进行Morris水迷宫实验观察其学习记忆能力,行为学检测后再通过免疫组化SABC法检测各组大鼠海马IL-6表达水平。结果模型组大鼠较对照组学习记忆能力降低,海马IL-6表达明显增强;治疗组大鼠较模型组学习记忆能力显著提高,海马IL-6表达显著下调。结论灵芝多糖可提高Aβ25-35诱导的模拟AD学习记忆障碍大鼠的学习记忆能力,抑制海马IL-6表达。
Objective To investigate the effect of Ganoderma lucidum polysaccharides on the expression of interleukin-6 (IL-6) in the brain of rats with simulated learning and memory impairment of AD. Methods A rat model of simulated learning and memory impairment was made by a one-time injection of β-amyloid peptide 25-35 fragment (Aβ25-35) in both hippocampus. 24 hours after the model was established, the treatment group received intraperitoneal injection of Ganoderma lucidum polysaccharide injection, and the other groups were respectively The same volume of normal saline was injected once a day for 7 days. Morris water maze experiment was performed on the 8th day to observe the learning and memory abilities. After the behavior test, the hippocampal IL-1 was detected by immunohistochemical SABC method. 6 expression levels. RESULTS: Compared with the control group, the rats in the model group had lower learning and memory ability and significantly increased IL-6 expression in the hippocampus. The rats in the treatment group had significantly higher learning and memory abilities than the model group, and the IL-6 expression in the hippocampus was significantly downregulated. Conclusion Ganoderma lucidum polysaccharides can improve learning and memory ability induced by Aβ25-35 in rats with simulated learning and memory impairment and inhibit the expression of IL-6 in hippocampus.
