目的研究硝酸甘油(GTN)抗柯萨奇B组3型病毒(CVB3)的作用特点和机制。方法比较先经GTN预处理及与GTN同时加入的两种处理方法相同滴度的病毒所引起的CPE抑制率的差异;分别测定并比较多次给予GTN及不同时间点给予GTN对CVB3感染的HeLa细胞组间的细胞病变效应抑制效果;测定GTN在HeLa细胞中不同时间点代谢产生的NO浓度,并做NO浓度与不同时间点给予GTN的细胞病变效应抑制结果的相关性分析。结果预加GTN组与同时加GTN组经等量病毒攻击所引起的CPE无显著性差异(P>0.05);多次给药的组间CPE抑制率无显著性差异(P>0.05),但不同时间点给药的组间CPE抑制率具有显著差异(P<0.05),NO浓度与不同时间点给予GTN的细胞病变效应抑制结果呈正相关(r=0.97,P<0.01)。结论GTN是通过其代谢产物NO来发挥抗CVB3作用的。
Objective To investigate the effect and mechanism of nitroglycerin (GTN) against Coxsackie B virus type 3 (CVB3). Methods The differences in CPE inhibition rate caused by the virus with the same titer as the GTN pretreatment and the two treatments simultaneously added with GTN were compared. The levels of CPE inhibition induced by GTN and CVB3 infection at different time points were measured and compared. The effect of GTN on NO production in HeLa cells at different time points was determined and the relationship between NO concentration and the inhibition of cytopathic effect of GTN at different time points was analyzed. Results There was no significant difference (P> 0.05) in the CPE between GTN group and GTN group (P> 0.05). There was no significant difference in the CPE inhibition rate between the two groups (P> 0.05) There was a significant difference (P <0.05) in the rate of CPE inhibition between the groups administered at different time points. The concentration of NO was positively correlated with the inhibition of cytopathic effect of GTN at different time points (r = 0.97, P <0.01). Conclusion GTN exerts anti-CVB3 effect through its metabolite NO.
