目的研究伤寒杆菌诱导的肠道黏膜免疫模型小鼠肠系膜淋巴结(MLN)内高内皮微静脉(HEVs)蛋白多糖(PG)的表达,探讨其对黏膜免疫淋巴细胞归巢的调控作用。方法采用阳性胶体铁染色光镜和透射电镜观察。结果蛋白多糖在伤寒杆菌诱导的黏膜免疫模型小鼠肠系膜淋巴结高内皮微静脉内皮细胞管腔面细胞膜呈强阳性表达。电镜下可见高内皮微静脉管腔内的淋巴细胞体积大小不等,多数细胞胞体较大,胞浆增多,细胞器丰富,淋巴细胞多呈活化状态。此外,还可见到多个淋巴细胞位于内皮细胞内、内皮细胞连接的间隙内以及基膜内,呈穿越状态。结论蛋白多糖对淋巴细胞穿越高内皮微静脉管壁以及向淋巴组织迁移可能有重要的调节作用。
Objective To investigate the expression of proteoglycan (PG) in the high endothelial cells (VECs) of mesenteric lymph node (MLN) in mice induced by Salmonella typhi and its regulation on the homing of mucosal immune lymphocytes. Methods Positive colloidal iron staining and transmission electron microscopy were used. Results Proteoglycan showed a strong positive expression in the membrane of the luminal surface of the endothelial cells in the mesenteric lymph nodes of mice with typhoid-induced mucosal immunity. Electron microscopy showed that the volume of lymphocytes in the lumen of the high-endothelial venules varied. Most of the cells had large cell bodies, increased cytoplasm, abundant organelles, and mostly activated lymphocytes. In addition, you can also see a number of lymphocytes located in endothelial cells, endothelial cells connected to the gap and the basement membrane, was through the state. Conclusion Proteoglycan may play an important regulatory role on the migration of lymphocytes through the walls of the high endothelial venules and into the lymphoid tissues.
