目的探讨沉默生殖器形成抑制基因-1(SMG-1)对人绒毛膜癌(人绒癌)JAR细胞凋亡以及转录激活子3(Stat3)表达作用的影响。方法实验分三组:空白对照组、阴性对照组和SMG-1 siRNA转染组。采用荧光定量PCR检测SMG-1 mRNA和Stat3 mRNA的相对表达水平,采用Western Blot方法检测转染前后Stat3和p-Stat3的蛋白表达水平,采用流式细胞术方法检测细胞凋亡情况。结果抑制SMG-1表达后,与空白对照组和阴性对照组相比,SMG-1 mRNA转染组SMG-1 siRNA和Stat3 mRNA相对表达水平均明显降低,差异有统计学意义(P<0.05);Stat3和p-Stat3蛋白表达水平均明显降低,差异有统计学意义(P<0.05),细胞凋亡率显著升高,差异有统计学意义(P<0.05)。结论沉默SMG-1可使Stat3和p-Stat3表达下降,细胞凋亡水平升高。
Objective To investigate the effects of silenced genital suppressor gene-1 (SMG-1) on the apoptosis and the expression of activator of transcription 3 (STAT3) in human choriocarcinoma (human choriocarcinoma) JAR cells. Methods The experiment was divided into three groups: blank control group, negative control group and SMG-1 siRNA transfection group. The relative expression levels of SMG-1 mRNA and Stat3 mRNA were detected by real-time PCR. The protein levels of Stat3 and p-Stat3 before and after transfection were detected by Western Blot. The apoptosis was detected by flow cytometry. Results Compared with the blank control group and the negative control group, the relative expression levels of SMG-1 siRNA and Stat3 mRNA in the SMG-1 mRNA transfected group were significantly decreased after SMG-1 expression was inhibited (P <0.05) ; Stat3 and p-Stat3 protein expression levels were significantly lower, the difference was statistically significant (P <0.05), the apoptosis rate was significantly increased, the difference was statistically significant (P <0.05). Conclusion Silencing SMG-1 can down-regulate the expression of Stat3 and p-Stat3 and increase the level of apoptosis.
