多种慢性肾病最后都会进展到终末期肾病 (end stagerenaldisease ,ESRD) ,其主要形态学特征是肾小球及肾小管间质纤维化。在此过程中 ,转化生长因子 β(transforminggrowthfactor β ,TGF β)被认为是致纤维化的关键因子。而结缔组织生长因子 (connectivetissuegrowthfactor,CTGF)是TGF β下游效应因子 ,它能介导TGF β的促细胞增生和细胞外基质 (extracellularmatrix ,ECM)合成。而近年来又发现 ,Rho蛋白在CT GF、TGF β基因调控中起关键性作用。本文重点讨论Rho蛋白的结构和功能、CTGF的结构与功能、Rho蛋白与CTGF、TGF β的关系及在肾小管间质纤维化中的作用、CTGF与TGF β的关系及CTGF在肾脏纤维化中的作用等。
A variety of chronic kidney diseases eventually progress to end stage renal disease (ESRD), the main morphological characteristics of glomerular and tubulointerstitial fibrosis. In the process, transforming growth factor β (TGFβ) is considered as a key factor for fibrosis. Connective tissue growth factor (CTGF) is a downstream factor of TGFβ, which can mediate the promotion of TGFβproliferation and extracellular matrix (ECM) synthesis. In recent years, it has been found that Rho protein plays a key role in the regulation of CTGF and TGFβgene. This article focuses on the structure and function of Rho protein, the structure and function of CTGF, Rho protein and CTGF, TGF β and its role in tubulointerstitial fibrosis, CTGF and TGF β and CTGF in renal fibrosis Role and so on.
