本研究以肺癌A549细胞株为实验材料,研究肿瘤坏死因子(TNF)对A549细胞单层及细胞球体的抑制作用,同时观察了TNF对A549细胞周期的影响.多细胞肿瘤球体(multicellular spheroid)在体外培养环境中的生长类似于实体瘤结节,具有活体肿瘤组织的许多特证,因此,利用多细胞球体研究药物对肿瘤细胞的抑制作用,能更好地反映药物及细胞因子的抗肿瘤效应.A549细胞体外转管培养1天,取直径为250μm大小的球体,转移至琼脂铺底的24孔细胞培养板中,每孔一个球体,分组加入TNF(10~4U/ml)及细胞培养液,继续培养,每天观察球体变化,测量相互垂直的两个长轴(a,b),用ab~(1/2)表示球体的平均直径.
In this study, lung cancer cell line A549 was used as experimental material to study the inhibitory effect of tumor necrosis factor (TNF) on monolayers and spheroids of A549 cells. The effect of TNF on the cell cycle of A549 cells was also observed. Multicellular spheroids were used in this study. Growth in an in vitro culture environment is similar to solid tumor nodules and has many special features of living tumor tissues. Therefore, the use of multicellular spheroids to study the inhibitory effect of drugs on tumor cells can better reflect the anti-tumor effects of drugs and cytokines. A549 cells were cultured in vitro for 1 day, and the spheres with a diameter of 250 μm were collected and transferred to a 24-well agar plate with one sphere per well. TNF (10~4 U/ml) and cell culture medium were added in groups. Continue training. Observe the changes of the sphere every day. Measure the two long axes (a, b) perpendicular to each other. Use ab (1/2) to represent the average diameter of the sphere.
