目的:探讨正常及高葡萄糖环境下,Fe2+对人脐静脉平滑肌细胞(HUVSMC)增殖及基质金属蛋白酶-2(MMP-2)分泌的作用。方法:以相同浓度接种普通培养液及高浓度葡萄糖培养液培养的3~5代HUVSMC为正常组及高糖组,分别以不同浓度Fe2+处理。培养24h后,四甲基氮唑盐(MMT)比色法测定细胞增殖水平,ELISA试剂盒检测细胞培养上清中MMP-2含量。结果:不同浓度葡萄糖环境下,0.0125~0.1000g/L浓度Fe2+均能促进HUVSMC增殖,且高糖组作用较正常组强;正常组Fe2+浓度在0.1g/L时能促进HUVSMC分泌MMP-2,高糖组所有浓度Fe2+均表现促分泌作用。结论:适当浓度的Fe2+能影响HUVSMC的生物学特性,促进HUVSMC的增殖与MMP-2的分泌,且高糖环境下促进作用更为显著。
AIM: To investigate the effect of Fe2 + on the proliferation and the secretion of matrix metalloproteinase-2 (MMP-2) in human umbilical vein smooth muscle cells (HUVSMCs) under normal and high glucose conditions. Methods: HUVSMCs of 3 to 5 generations cultured in the same culture medium and high glucose medium at the same concentration were treated with different concentrations of Fe2 +, respectively. After cultured for 24h, the cell proliferation was measured by MTT colorimetric assay. The content of MMP-2 in the cell culture supernatant was detected by ELISA kit. Results: Under the different concentrations of glucose, Fe2 + concentration of 0.0125-0.1000g / L could promote the proliferation of HUVSMC, and the effect of high glucose group was stronger than that of normal group. When Fe2 + concentration was 0.1g / L, the secretion of MMP- All concentrations of Fe2 + in high glucose group showed secreting effect. Conclusion: Fe2 + of appropriate concentration can affect the biological characteristics of HUVSMC, promote the proliferation of HUVSMC and the secretion of MMP-2, and the promotion effect is more significant in high glucose environment.
