目的:通过观察丁香的3种提取物(挥发油、水提液、去挥发油水提液)对寒证大鼠脑内神经递质去甲肾上腺素(NE)、多巴胺(DA)、5-羟色胺(5-HT)、及环磷酸腺苷(cAMP)、环磷酸鸟苷(cGMP)的影响,探讨丁香3种提取物与温热药性的相关性,以及入煎剂的科学性。方法:设空白对照组、模型对照Ⅰ组、模型对照Ⅱ组、阳性对照组、丁香3种提取物高、低剂量组,用寒凉药复制大鼠寒证模型,荧光光度法测定NE,DA的含量,酶联法测定5-HT的含量;放免法测定cAMP,cGMP的含量。结果:丁香水提液高剂量、挥发油高、低剂量、去挥发油水提液高剂量组均能增加NE的含量(P<0.05或P<0.01)。丁香水提液与挥发油高、低剂量组,去挥发油水提液高剂量组均能增加DA的含量(P<0.05或P<0.01),减少5-HT的含量(P<0.05或P<0.01);丁香水提液与去挥发油高剂量组,挥发油高、低剂量组均能增加cAMP的含量(P<0.05或P<0.01),升高cAMP/cGMP比值(P<0.05或P<0.01),但对cGMP没有显著影响。结论:丁香不同提取物能促进寒证大鼠脑内NE,DA的合成,抑制5-HT的释放,通过促进大脑皮层中cAMP的合成,调节大脑皮层中cAMP和cGMP的平衡;丁香挥发油与其温热药性存在相关性,而水提液、去挥发油水提液部分也存在与温热药性相关的活性成分,以煎剂入药具有科学性。
OBJECTIVE: To observe the effects of three extracts of clove (volatile oil, water extract and debridement oil extract) on the neurotransmitters NE, DA and 5-HT 5-HT, cAMP and cGMP, and to explore the correlation between the three extracts of clove and the warming medicinal properties and the feasibility of decoction. Methods: Cold and cold medicine were used to establish the model of cold syndrome in model control group Ⅰ, model control group Ⅱ, positive control group and clove three high and low dose groups. The levels of NE and DA were determined by fluorophotometry The content of 5-HT was determined by enzyme-linked immunosorbent assay. The levels of cAMP and cGMP were determined by radioimmunoassay. Results: The results showed that the content of NE increased (P <0.05 or P <0.01) by high dose clove water extract, high and low dose of volatile oil, high dose debridement and water extract. L clove water extract and volatile oil high, low dose group, high volatile oil extract group can increase the content of DA (P <0.05 or P <0.01), reduce the content of 5-HT (P <0.05 or P <0.01 ), Clove water extract and de-oiling high-dose group, volatile oil high-dose and low-dose group can increase the content of cAMP (P <0.05 or P <0.01), increase the cAMP / cGMP ratio (P <0.05 or P <0.01) , But no significant effect on cGMP. CONCLUSION: Different extracts of clove can promote the synthesis of NE and DA, inhibit the release of 5-HT and improve the balance of cAMP and cGMP in cerebral cortex by promoting cAMP synthesis in the cerebral cortex. There is a correlation between the thermo-medicinal properties, while the aqueous extract, to the volatile oil extract part also exists with the active ingredient of the thermo-medicinal properties, to decoction medicine is scientific.
