目的:观察复方三七纳米颗粒对脑创伤后大鼠血脑屏障、肿瘤坏死因子-α(TNF-α)和白介素-6(IL-6)的影响作用。方法:用Feeney自由落体撞击法建立成年雄性大鼠脑创伤模型,96只实验动物随机分为空白对照组、创伤模型组、复方三七纳米颗粒低、高剂量组(0.6,2.4 g·kg-1),每组24只大鼠。第1,3,5天参照ZeaLonga法,对各组实验大鼠进行神经功能缺损评分;之后经腹主动脉穿刺取血,用双抗体夹心酶联免疫吸附法(ELISA)测量各组不同时间点大鼠(n=8)血清肿瘤坏死因子-α(TNF-α)和白介素-6(IL-6)含量的变化;之后处死大鼠,取造模损伤部位脑组织(约4 mm×4 mm×3 mm)称重计算脑组织含水量,观察用药后各时间点大鼠脑水肿情况。结果:与空白对照组比较,模型组神经功能缺损评分升高,脑组织含水量、TNF-α和IL-6含量均增加;与模型组比较,复方三七纳米颗粒干预组中神经功能缺损评分降低,脑组织含水量、TNF-α和IL-6含量均降低,并且随着剂量的增加上述变化显著(P<0.05)。结论:复方三七纳米颗粒对大鼠脑创伤后有保护作用,与TNF-α的含量降低和IL-6含量增加有关。
Objective: To observe the effect of Panax notoginseng nanoparticle on the blood-brain barrier, tumor necrosis factor-α (TNF-α) and interleukin-6 (IL-6) in rats after traumatic brain injury. Methods: Adult male rat brain trauma model was established by Feeney free-fall impact method. 96 experimental animals were randomly divided into blank control group, trauma model group, low and high dose Panax notoginseng nanoparticles group (0.6, 2.4 g · kg- 1), 24 rats per group. On the 1st, 3rd and 5th days, the neurological deficits were evaluated by ZeaLonga method. The blood samples were taken from the abdominal aorta, and the blood samples were collected by double antibody sandwich enzyme-linked immunosorbent assay (ELISA) The levels of tumor necrosis factor-α (TNF-α) and interleukin-6 (IL-6) in serum of rats (n = 8) × 3 mm) Weighing Calculate the water content of brain tissue and observe the brain edema in rats at each time point after administration. Results: Compared with the blank control group, the neurological deficit score of the model group increased, and the content of water, TNF-α and IL-6 in brain tissue increased. Compared with the model group, the neurological deficit score Decreased, water content in brain tissue, TNF-α and IL-6 decreased, and the above-mentioned changes were significant with the increase of dosage (P <0.05). Conclusion: Panax notoginseng nanoparticles have a protective effect on traumatic brain injury in rats, which is related to the decrease of TNF-α content and the increase of IL-6 content.
