荔枝核皂苷对高糖诱导的HMC细胞增殖和凋亡的作用

目的探讨高糖环境下人肾小球系膜细胞(HMC)的增殖及荔枝核皂苷的干预作用,阐明荔枝核皂苷对高糖诱导的HMC增殖的作用机制。方法体外培养HMC细胞,给予高浓度葡萄糖(30 mmol/L)诱导HMC细胞,加入不同浓度的荔枝核皂苷进行干预处理。应用四甲基偶氮唑蓝(MTT)法检测高糖及不同浓度荔枝核皂苷干预HMC细胞的增殖情况;ELISA法检测转化生长因子(TGF-β1)和纤维连接蛋白(FN)的含量;流式Annexin V-FITC/PI双染法检测细胞凋亡情况。结果 MTT法及ELISA法检测结果显示:荔枝核皂苷各剂量组均可抑制高糖引起的HMC细胞增殖(P<0.05或P<0.01),并降低TGF-β1、FN的含量。流式细胞术检测结果显示:荔枝核皂苷各剂量组,48h细胞凋亡率表达明显增加(P<0.01)。结论荔枝核皂苷对高糖诱导的HMC细胞增殖具有抑制作用,并促进其凋亡,其抑制作用可能是通过促进细胞凋亡的途径抑制细胞增殖,此作用可能对延缓和控制糖尿病肾病有积极作用。 Objective To investigate the proliferation of human glomerular mesangial cells (HMC) and the intervention of lychee nuclear saponin in high glucose environment and to elucidate the mechanism of lychee nuclear saponins on the proliferation of HMC induced by high glucose. Methods HMC cells were cultured in vitro. HMC cells were induced by high concentration glucose (30 mmol / L), and different concentrations of lychee nucleosides were intervened. The proliferation of HMC cells treated with high glucose and different concentrations of litchi were detected by MTT assay. The levels of TGF-β1 and fibronectin (FN) were detected by ELISA. Apoptosis was detected by Annexin V-FITC / PI double staining. Results The results of MTT assay and ELISA assay showed that each dose of litchi could inhibit the proliferation of HMC cells induced by high glucose (P <0.05 or P <0.01) and decrease the content of TGF-β1 and FN. The results of flow cytometry showed that the expression of apoptosis rate increased significantly at 48h (P <0.01) in each dose of litchi nucleoside. Conclusion Lychee Saponins can inhibit the proliferation of HMC induced by high glucose and promote its apoptosis. The inhibitory effect may be due to the inhibition of cell proliferation by promoting apoptosis, which may have a positive effect on delaying and controlling diabetic nephropathy .
