【目的】优选土茯苓多糖硫酸化修饰的最佳条件,探讨硫酸化修饰提高土茯苓多糖活性的可能性。【方法】采用氯磺酸—吡啶法对纯化的土茯苓多糖进行硫酸化修饰,以产率、硫酸基取代度、糖含量作为考察指标,用L9(34)正交实验设计对反应时间、反应温度和氯磺酸—吡啶配比3个因素进行优化,用氯化钡—明胶法测定硫酸基的取代度(DS),用硫酸—苯酚法测定糖含量,红外光谱测定其结构,并用噻唑蓝(MTT)比色法检测正交试验中的各种硫酸化后多糖产物对肿瘤细胞Hep G2和MCF-7增殖活性的影响。【结果】土茯苓多糖采用氯磺酸—吡啶法进行硫酸化修饰的最佳条件为反应时间4.5 h,反应温度60℃和试剂配比为1∶6;MTT结果显示:硫酸化后的多糖产物对肿瘤细胞Hep G2和MCF-7具有一定的抑制增殖作用,并与浓度呈依赖性关系。【结论】氯磺酸—吡啶法对土茯苓多糖进行硫酸化修饰是可行的,且硫酸化修饰能提高土茯苓多糖的抗肿瘤活性。
【Objective】 The optimum conditions for sulfation of Smilax glabra were optimized and the possibility of sulfation modification to improve the activity of Smilax Polysaccharide was discussed. 【Method】 Purified Smilax glabra polysulfate was sulfated with chlorosulfonic acid-pyridine method. The yield, the degree of substitution of sulfonic acid group and the content of sugar were taken as the indexes of investigation. The reaction time, reaction time Temperature and the ratio of chlorosulfonic acid to pyridine were optimized. The degree of substitution (DS) was determined by barium chloride-gelatin method. The content of carbohydrate was determined by sulfuric acid-phenol method. The structure was determined by infrared spectroscopy. (MTT) assay was used to detect the effects of various sulfated polysaccharides in the orthogonal test on the proliferation of Hep G2 and MCF-7 tumor cells. 【Result】 The results showed that the optimal conditions for the sulfation of Smilax glabra polysaccharides by chlorosulfonic acid-pyridine method were as follows: reaction time 4.5 h, reaction temperature 60 ℃ and reagent ratio 1: 6. MTT results showed that sulfated polysaccharide product Hep G2 and MCF-7 have certain inhibitory effect on proliferation, and have a concentration-dependent relationship. 【Conclusion】 Chlorosulfonic acid-pyridine method is feasible for sulfation of Smilax glabra polysaccharides, and sulfation modification can increase the anti-tumor activity of Smilax Polysaccharide.
