目的:研究红活麻总香豆素对葡聚糖硫酸钠(DSS)所致结肠炎小鼠的防治作用。方法:采用DSS诱发小鼠溃疡性结肠炎模型,给予红活麻总香豆素后,观察结肠炎小鼠体重变化,ELISA检测血清IL-6,IL-10,TGF-β1,IFN-γ的水平,分离结肠组织进行病理学检查,Western blot检测结肠组织TLR4和NF-κB的表达水平。结果:红活麻总香豆素能有效控制结肠炎小鼠体重下降;降低血清IL-6,IFN-γ,升高抑制性细胞因子IL-10,TGF-β1水平;减少结肠组织损伤,降低结肠组织TLR4和NF-κB的表达水平。结论:红活麻总香豆素通过调节促炎/抗炎细胞因子的水平,降低结肠组织TLR4和NF-κB的表达,发挥抗小鼠结肠炎的作用。
Objective: To study the preventive and curative effect of the red coumarin coumarin on colitis mice induced by dextran sodium sulfate (DSS). Methods: The model of ulcerative colitis in mice was induced by DSS. The weight of colitis mice was observed after administration of total coumarin. The levels of IL-6, IL-10, TGF-β1 and IFN-γ The colon tissues were isolated and pathologically examined. Western blot was used to detect the expression of TLR4 and NF-κB. Results: The total coumarin could inhibit the body weight of colitis mice, reduce the levels of serum IL-6 and IFN-γ, increase the levels of inhibitory cytokines IL-10 and TGF-β1, reduce the damage of colonic tissue and decrease Colonic Tissue TLR4 and NF-κB expression levels. CONCLUSION: CTZ can reduce the expression of TLR4 and NF-κB in colonic tissue by regulating the levels of proinflammatory and anti-inflammatory cytokines and exert anti-mouse colitis.
