目的:分析喉癌患者与健康人血浆中差异表达的蛋白质组,筛选潜在的与喉癌患者诊断密切相关的血浆分子标志物。方法:取喉癌患者与健康人空腹血浆标本各6例,分别等量混合成2组样本,利用双向电泳(2-DE)及基质辅助激光解吸电离飞行时间质谱(MALDI-TOF/TOF)技术筛选及鉴定差异表达的蛋白点,并对其进行生物信息学分析。结果:喉癌患者与健康人血浆相比较,筛选出表达量差异1.5倍以上的差异蛋白点共20个,其中表达上调的有8个,表达下调的有12个,进行质谱分析后,表达上调的蛋白有α-1抗胰蛋白酶、α-1酸性糖蛋白1、载脂蛋白L1、C反应蛋白、结合珠蛋白、血浆铜蓝蛋白;表达下调的蛋白有转铁蛋白、α-2-HS-糖蛋白、纤维蛋白原γ链、结合珠蛋白相关蛋白、免疫球蛋白λ-1链C结构域、免疫球蛋白κ链C结构域、载脂蛋白A1、转甲状腺素蛋白、载脂蛋白C3。结论:喉癌患者与健康人血浆的蛋白质表达谱表现出一定差异,这些差异蛋白有可能成为喉癌患者诊断的特异性血浆肿瘤标志物。
OBJECTIVE: To analyze differentially expressed proteome in the plasma of patients with laryngeal cancer and healthy people and to screen potential plasma molecular markers closely related to the diagnosis of laryngeal cancer patients. Methods: Six patients with laryngeal cancer and healthy volunteers were divided into two groups according to the same dose. Two-dimensional electrophoresis (2-DE) and matrix-assisted laser desorption / ionization time of flight mass spectrometry (MALDI-TOF / TOF) Screening and identification of differentially expressed protein spots, and its bioinformatics analysis. Results: Compared with the plasma of healthy people, the number of differentially expressed protein spots in the laryngeal cancer patients was more than 1.5 times of the normal ones, of which 20 were up-regulated, 8 were down-regulated, 12 were down-regulated, and were upregulated after mass spectrometry analysis Α-1 antitrypsin, α-1 acid glycoprotein 1, apolipoprotein L1, C-reactive protein, haptoglobin, ceruloplasmin; down-regulated protein transferrin, α-2-HS - glycoprotein, fibrinogen gamma chain, haptoglobin related protein, immunoglobulin λ-1 chain C domain, immunoglobulin κ chain C domain, apolipoprotein A1, transthyretin, apolipoprotein C3 . Conclusion: The protein expression profiles of plasma in laryngeal cancer patients and healthy individuals show some differences. These differential proteins may become specific plasma tumor markers for diagnosis in patients with laryngeal cancer.
