目的:观察活血通督汤对脊髓缺血再灌注损伤(SCII)脊髓组织肿瘤坏死因子(TNF-α)、白细胞介素(IL)-1β、IL-8表达的影响,探讨其作用与机制。方法:72只新西兰家兔随机分为假手术组(n=8)、模型组(n=32)、活血通督汤组(n=32)。模型组、活血通督汤组采用夹闭腰动脉法制作SCII模型。假手术组不夹闭腰动脉,其它步骤同模型组。采用免疫组化法检测脊髓缺血再灌注后0.5、1、4、8h后脊髓组织中TNF-α的表达情况,ELISA法检测各观测时点IL-1β、IL-8的表达情况。结果:假手术组TNF-α、IL-1β、IL-8表达较低;模型组和活血通督汤组TNF-α、IL-1β、IL-8在0.5h即开始增高,并随再灌注的时间延长而持续增高;与模型组比较,活血通督汤组各时间点TNF-α、IL-1β、IL-8的表达显著减少(P<0.05,P<0.01)。结论:活血通督汤作用机制之一是通过减少脊髓组织的TNF-α、IL-1β、IL-8表达,抑制炎性反应,从而减轻SCII。
Objective: To observe the effect of Huoxuetongdu decoction on the expression of tumor necrosis factor (TNF-α), interleukin (IL) -1β and IL-8 in spinal cord after spinal cord ischemia-reperfusion injury (SCII), and to explore its mechanism. Methods: Seventy - two New Zealand rabbits were randomly divided into sham operation group (n = 8), model group (n = 32) and Huoxuetongsuotang group (n = 32). Model group, Huoxuetuotang group occlusion of lumbar artery method to make SCII model. The sham operation group did not clamp the lumbar artery, the other steps were the same as the model group. Immunohistochemistry was used to detect the expression of TNF-α in the spinal cord at 0.5, 1, 4 and 8 hours after spinal cord ischemia / reperfusion. The expression of IL-1β and IL-8 at each observation time was detected by ELISA. Results: The expression of TNF-α, IL-1β and IL-8 in sham operation group was lower than that in sham operation group. TNF-α, IL-1β and IL-8 in model group and Huoxutongsuotang group began to increase at 0.5h, (P <0.05, P <0.01). Compared with the model group, the expression of TNF-α, IL-1β and IL-8 in Huoxutongsuotang group was significantly decreased at each time point (P <0.05, P <0.01). Conclusion: One of the mechanisms of action of Huoxue Tongsu Tang is to reduce SCII by decreasing the expression of TNF-α, IL-1β and IL-8 in the spinal cord and inhibiting the inflammatory reaction.
