目的:评价氟康唑和他克莫司联用对光滑念珠菌耐药基因表达量的影响。方法:采用实时定量PCR定性筛查联用药物对各耐药基因氟康唑耐药基因ERG11,CDR1,PDH1和SNQ2表达水平的影响,并用RT-q PCR定量检测各耐药基因在不同药物处理下的表达量。结果:与对照组相比,氟康唑和他克莫司联用使ERG11和SNQ2基因的表达量分别减少了90%和66%,CDR1基因的表达量则增加了311.6%(P<0.05或P<0.01),对PDH1基因的表达无显著影响(P>0.05)。结论:他克莫司和氟康唑联合用药可能是克服光滑念珠菌对氟康唑耐药性的方法。
Objective: To evaluate the effect of fluconazole and tacrolimus on the expression of Candida glabrata-resistant gene. Methods: The real-time quantitative PCR was used to qualitatively screen the expression of fludazole resistance gene ERG11, CDR1, PDH1 and SNQ2 in each drug resistance gene. The quantitative RT-q PCR was used to detect the expression of different drug resistance genes in different drugs Under the expression level. Results: Fluconazole combined with tacrolimus reduced the expression of ERG11 and SNQ2 by 90% and 66%, respectively, and increased the expression of CDR1 by 311.6% (P <0.05 or P <0.01), but no significant effect on PDH1 gene expression (P> 0.05). Conclusion: The combination of tacrolimus and fluconazole may be the way to overcome the resistance of Candida glabrata to fluconazole.
