目的研究赤芍总苷(TSP)对实验性血栓形成的影响及其作用机制。方法(1)采用体外血栓法,观察赤芍总苷对体外血栓长度和干、湿重的影响;(2)建立血瘀模型,观察赤芍总苷对血瘀大鼠血液黏度、红细胞比容、血小板聚集率和凝血时间的影响。结果(1)赤芍总苷20.0和40.0 mg.kg-1可缩短体外血栓长度(P<0.05,P<0.01),降低体外血栓湿重和干重(P<0.05,P<0.01);(2)赤芍总苷20.0和40.0 mg.kg-1可明显降低红细胞比容、血液黏度(P<0.05,P<0.01),降低血小板聚集率,延长凝血时间(P<0.05,P<0.01)。结论赤芍总苷具有抗血栓作用,其机制与保护血管内皮细胞损伤,降低血液黏度,改善血液流变性有关。
Objective To investigate the effect of total glucosides of radix paeoniae (TSP) on experimental thrombosis and its mechanism of action. Methods (1) In vitro thrombosis method was used to observe the effects of total glucosides of Radix Paeoniae Rubra on in vitro thrombus length and dry and wet weights; (2) Blood Stasis model was established to observe the blood viscosity and hematocrit of blood total glycosides in rats. The effect of platelet aggregation rate and clotting time. Results (1) Total glucosides 20.0 and 40.0 mg.kg-1 decreased the in vitro thrombus length (P<0.05, P<0.01) and decreased the thrombus wet weight and dry weight in vitro (P<0.05, P<0.01); 2) Total glucosides 20.0 and 40.0 mg.kg-1 significantly decreased hematocrit, blood viscosity (P<0.05, P<0.01), decreased platelet aggregation rate, and prolonged clotting time (P<0.05, P<0.01). . Conclusion The total glucosides of Radix Paeoniae Radix has the antithrombotic effect. The mechanism is related to the protection of vascular endothelial cell injury, the decrease of blood viscosity and the improvement of hemorheology.
