目的:探讨黄连解毒汤及其主要成分小檗碱、黄芩苷、巴马汀、栀子苷对过氧化氢(H2O2)、谷氨酸(Glu)所致神经细胞损伤的影响。方法:制备H2O2,Glu损伤人神经母细胞瘤细胞(SH-SY5Y)、小鼠神经母细胞瘤细胞(Neuro-2a)模型,设正常对照、模型、黄连解毒汤、小檗碱、黄芩苷、巴马汀及栀子苷组,筛选浓度为7.812 5,15.625,31.25,62.5,125,250,500 mg.L-1,应用MTT法检测药物对损伤后细胞增殖的影响。结果:黄连解毒汤能够促进SH-SY5Y细胞增殖,适当浓度的栀子苷能够促进SH-SY5Y,Neuro-2a细胞增殖,并对H2O2,Glu损伤的SH-SY5Y细胞有保护作用,对H2O2损伤的Neuro-2a也具有保护作用。结论:栀子苷可能为黄连解毒汤对神经细胞保护作用的有效成分。
Objective: To investigate the effects of berberine, baicalin, palmatine and geniposide on the injury of neurons induced by hydrogen peroxide (H2O2) and glutamate (Glu) of Huanglian Jiedu Decoction and its main components. Methods: H2O2, Glu-induced human neuroblastoma cells (SH-SY5Y) and mouse neuroblastoma cells (Neuro-2a) model were established. Normal control, model, berberine, berberine, Palmatine and geniposide group. The screening concentration was 7.812 5,15.625,31.25,62.5,125,250,500 mg.L-1. The effect of drugs on cell proliferation was detected by MTT assay. Results: Huanglian Jiedu Decoction could promote the proliferation of SH-SY5Y cells. Appropriate concentration of geniposide could promote the proliferation of SH-SY5Y and Neuro-2a cells and protect SH-SY5Y cells induced by H2O2 and Glu. Neuro-2a also has a protective effect. Conclusion: Geniposide may be an effective component of Huanglian Jiedu Decoction on nerve cell protection.
