目的:探讨胃癌患者体内氧化及抗氧化水平的变化与胃癌的关系。方法:以胃腺癌患者(n=48)和健康人(n=30)空腹外周血为标本,采用紫外分光光度法检测氧化酶指标(MPO、XOD)、总抗氧化能力(T-AOC)以及丙二醛(MDA)的表达水平,采用酶联免疫吸附法(ELISA)测定8-羟基鸟嘌呤(8-OHdG)的表达。采用荧光定量RT-PCR法测定胃腺癌组织(n=68)和正常组织(n=12)中8-羟基鸟嘌呤糖苷酶mRNA表达水平。结果:胃癌组血浆XOD、MPO表达水平分别为(10.46±7.23)U/L、(23.71±25.06)U/L,健康对照组血浆XOD、MPO表达水平分别为(5.85±2.27)、(5.16±11.35)U/L,胃癌组均高于对照组,差异具有统计学意义(P<0.05);胃癌组T-AOC的表达水平为(5.02±3.13)U/mL,健康对照组为(19.40±7.18)U/mL,胃癌组低于对照组(P<0.05);胃癌组血浆8-OHdG和MDA的表达水平分别为(14.76±12.41)ng/mL、(18.83±4.19)nmol/mL,健康对照组为(6.49±5.75)ng/mL、(7.11±6.20)nmol/mL,胃癌组高于对照组(P<0.05)。胃癌组织中hOGG1 mRNA相对表达值(5.34±2.37)高于对照组的表达(1.04±0.76),差异有统计学意义(Z=1.964,P<0.05)。结论:胃腺癌患者体内氧化应激水平升高以及抗氧化防御水平的降低,胃腺癌组织中hOGG1mRNA表达增强。
Objective: To investigate the relationship between gastric cancer and the changes of oxidation and anti-oxidation in gastric cancer patients. Methods: The fasting peripheral blood samples of patients with gastric adenocarcinoma (n = 48) and healthy people (n = 30) were collected. The oxidative stress index (MPO, XOD), total antioxidant capacity (T-AOC) The expression of 8-hydroxyguanine (8-OHdG) was detected by enzyme linked immunosorbent assay (ELISA). The levels of 8-oxoguanine glycosylase mRNA in gastric adenocarcinoma (n = 68) and normal tissues (n = 12) were determined by fluorescence quantitative RT-PCR. Results: The plasma levels of XOD and MPO in gastric cancer group were (10.46 ± 7.23) U / L and (23.71 ± 25.06) U / L, respectively. The levels of XOD and MPO in healthy controls were (5.85 ± 2.27) and (P <0.05). The expression of T-AOC in gastric cancer group was (5.02 ± 3.13) U / mL and that in healthy control group was (19.40 ± 11.35) U / L and gastric cancer group was significantly higher than that in control group 7.18) U / mL in gastric cancer group was lower than that in control group (P <0.05). The expression of 8-OHdG and MDA in gastric cancer group were (14.76 ± 12.41) ng / mL and (18.83 ± 4.19) nmol / The control group was (6.49 ± 5.75) ng / mL, (7.11 ± 6.20) nmol / mL, which was significantly higher in the gastric cancer group than in the control group (P <0.05). The relative expression value of hOGG1 mRNA in gastric cancer tissues (5.34 ± 2.37) was higher than that in the control group (1.04 ± 0.76), with significant difference (Z = 1.964, P <0.05). Conclusion: In patients with gastric adenocarcinoma elevated levels of oxidative stress and decreased levels of antioxidant defense, hOGG1mRNA expression in gastric adenocarcinoma increased.
