目的探讨中国圆田螺多糖体内抗鸭乙肝病毒(DHBV)作用。方法 PCR检测1d龄绍兴麻鸭血清DHBV-DNA,筛选出先天感染DHBV鸭30只,随机分为模型对照组,3TC对照组和中国圆田螺多糖高、中、低剂量组,每组6只,连续灌胃给药14d,每天2次,分别于给药前、给药7d、给药14d及停药后5d经腿胫静脉取血,分离血清,采用荧光探针定量PCR检测DHBV-DNA含量变化,采用酶联免疫吸附法检测DHBsAg和DHBeAg滴度变化。结果中国圆田螺多糖中、高剂量组在给药14d、停药5d时对DHBV-DNA均有明显抑制作用(t值分别为:15.46、17.90、26.92、24.51,P<0.01);中国圆田螺多糖中、高剂量组在给药14d及停药5d时对DHBsAg表达均有抑制作用(t值分别为:6.88、3.95、11.71、9.45,P<0.05);中国圆田螺多糖中、高剂量组在给药7d、14d及停药5d时对DHBeAg的表达亦有抑制作用(t值分别为:3.19、3.79、3.03、4.08、4.25、4.07,P<0.05);3TC对照组在给药7d和14d时对DHBV-DNA、DHBsAg和DHBeAg有明显抑制作用(P<0.01),但在停药5d时抑制作用显著下降出现反跳现象。结论中国圆田螺多糖能抑制鸭血清中DHBV-DNA、DHBsAg和DHBeAg的表达量,即具有体内抗DHBV活性。
Objective To investigate the anti-duck hepatitis B virus (DHBV) effect of polysaccharides from Crytomeria fortunei in China. Methods DHBV-DNA of Shaoxing geese aged 1 d was detected by PCR. Thirty DHBV congenital infections were selected and randomly divided into model control group, 3TC control group and high, medium and low dose of Chinese snail polysaccharide, 6 rats in each group. Serum was administered intragastrically for 14 d twice daily before administration, 7 d after drug administration, 14 d after drug administration and 5 d after drug withdrawal, respectively. The serum was separated and the content of DHBV-DNA was detected by fluorescent probe quantitative PCR Changes, the use of enzyme-linked immunosorbent assay DHBsAg and DHBeAg titer changes. Results The DHBV-DNA was significantly inhibited at 14 d and 5 d after drug administration (t = 15.46, 17.90, 26.92, 24.51, P <0.01, respectively) In polysaccharides and high dose groups, DHBsAg expression was inhibited at 14d and 5d (t = 6.88, 3.95, 11.71, 9.45 respectively, P <0.05) The expression of DHBeAg was also inhibited 7d, 14d and 5d after administration (t = 3.19,3.79,3.03,4.08,4.25,4.07, P <0.05) On the 14th day, DHBV-DNA, DHBsAg and DHBeAg were significantly inhibited (P <0.01), but the inhibition was significantly decreased at 5 days after drug withdrawal. CONCLUSION The polysaccharides from Cryptomeria fortunei can inhibit the expression of DHBV-DNA, DHBsAg and DHBeAg in duck serum, that is, have anti-DHBV activity in vivo.
