目的:了解兔源性透明质酸刺激因子(hyaluronicacidstimulatingfactor,HASF)对人增生性瘢痕成纤维细胞生长增殖的影响.方法:将HASF作用于体外培养的增生性瘢痕成纤维细胞,测定3H-TdR参入量、MTT反应A值及上清液中透明质酸(HA)的含量.结果:1×10-3~1×10-2g/LHASF可剂量依赖性地使细胞3H-TdR参入量及MTT反应A值降低,HA含量明显增加(P<0.01).结论:兔源性HASF在体外能促进人增生性瘢痕成纤维细胞合成HA,对其生长增殖有明显的抑制作用;这种抑制作用可能是通过HASF刺激细胞合成HA来实现的.
Objective: To investigate the effect of hyaluronic acid-stimulating factor (HASF) on proliferation and proliferation of human hypertrophic scar fibroblasts. Methods: HASF was used to treat hypertrophic scar fibroblasts in vitro. 3H-TdR incorporation, A-value of MTT reaction and content of HA in the supernatant were determined. Results: 1 × 10-3 ~ 1 × 10-2g / LHASF could decrease the 3H-TdR incorporation and the A value of MTT reaction in a dose-dependent manner, and the content of HA significantly increased (P <0.01). CONCLUSION: Rabbit-derived HASF can promote the synthesis of HA in human hypertrophic scar fibroblasts in vitro, which can significantly inhibit the growth and proliferation of HFCs. The inhibitory effect may be achieved by stimulating HASF cells to synthesize HA.
