目的:采用高效液相梯度洗脱法测定马鬃蛇药酒(MZSYJ)中染料木苷、染料木素、原儿茶酸和没食子酸的含量。方法:Hypersil C18色谱柱(4.6 mm×200 mm,5μm);流速1.0 mL·min-1。染料木苷和染料木素:流动相A为乙腈-甲醇(2∶1),流动相B为0.2%甲酸水溶液,检测波长261 nm。原儿茶酸和没食子酸:流动相A为甲醇,流动相B为0.3%磷酸溶液,检测波长268 nm。结果:染料木苷和染料木素分别在0.010 4~0.208 0μg(r=0.999 3),0.004 8~0.096 0μg(r=0.999 6)进样量与峰面积呈良好的线性关系,平均加样回收率分别为97.18%,96.94%,RSD分别为1.52%,1.40%;原儿茶酸和没食子酸分别在0.038 2~0.764 0μg(r=0.999 1),0.011 6~0.232 0μg(r=0.999 4)进样量与峰面积呈良好的线性关系,平均加样回收率分别为95.97%,98.39%,RSD分别为1.10%,1.45%。结论:该方法测定结果准确、灵敏、重复性好。
Objective: To determine the contents of genistin, genistein, protocatechuic acid and gallic acid in MZSYJ by high performance liquid chromatography gradient elution. METHODS: Hypersil C18 column (4.6 mm × 200 mm, 5 μm) was used. The flow rate was 1.0 mL · min-1. Genistin and genistein: mobile phase A was acetonitrile-methanol (2: 1), mobile phase B was 0.2% formic acid in water and the detection wavelength was 261 nm. Protocatechuic acid and gallic acid: mobile phase A methanol, mobile phase B 0.3% phosphoric acid solution, detection wavelength 268 nm. Results: The dosages of genistin and genistein were linearly correlated with the peak area in the range of 0.010 4-0.208 0 μg (r = 0.999 3) and 0.004 8-0.096 0 μg (r = 0.999 6), respectively. The mean sample recovery The relative standard deviations (RSDs) were 97.18% and 96.94%, respectively, with RSDs of 1.52% and 1.40%, respectively.
