将长2~3cm的小麦幼穗剪成2~3mm长的小切段接种于添加2,4-D、NAA的MS培养基上,当愈伤组织直径达5mm左右时,不加分割直接转移到分化培养基上培养,获得54个试管变态小穗,其颖片和小花柄明显伸长,呈簇生状或分枝状,比正常的大2~8倍,花器发育完全。在无菌条件下授以黑麦花粉,产生11粒杂交种子。经离体胚培养,获得8株试管幼苗。
The young 2 ~ 3cm long wheat ears were cut into small pieces of 2 ~ 3mm in length and inoculated on 2,4-D and NAA-supplemented MS medium.
