应用全细胞膜片钳技术,研究巴氯芬(baclofen)对新鲜分离的大鼠DRG神经元的作用。结果发现:GABAB受体选择性激动剂baclofen(10-5和10-4mol/L)对蝇蕈醇(muscimol)(10-5mol/L)和异四氢烟酸(isoguvacine)(5×10-5mol/L)-激活电流有抑制作用,并呈浓度依赖性。并对baclofen抑制GABAA选择性受体激动剂muscimol-和isoguvacine-激活电流反应可能的胞内转导机制进行了讨论。
Whole cell patch clamp techniques were used to study the effect of baclofen on freshly isolated rat DRG neurons. As a result, it was found that the effects of baclofen (10-5 and 10-4 mol / L) on muscimol (10-5 mol / L) and isoguvacine (5x10- 5mol / L) - activation current inhibition, and in a concentration-dependent manner. And discussed the possible intracellular transduction mechanism of baclofen inhibiting the GABAA selective receptor agonists muscimol- and isoguvacine-activated currents.
