5-FU、DDP既是有效的化疗药物又对辐射有增敏作用。作为增敏药物在其与放射合用时要达到最大的增敏效果,必然有最佳的放化时间顺序。为探讨此问题,我们采用离体细胞克隆培养技术,观察了5-Fu+DDP联合放射,在放射前、中、后不同时相给药5小时,对人肺腺癌Anip973细胞的影响。结果,3个放化结合组DEF值均大于1,细胞存活率比单照和单纯给药明显减低;先药后照组细胞杀灭倍数为4.57,大于给药中间照射组和先照后药组;DEF值以先药后照组为最高(2.2);细胞平均存活率,先药后照组低于其它两组,分别为0.096,0.125,0.115.本实验结果显示,5-Fu+DDP联合放射的增敏作用,在放射前给药并保持一定的药物作用时间,效果最好。
5-FU and DDP are both effective chemotherapeutic agents and have sensitizing effects on radiation. As the sensitizing drug has to achieve the maximum sensitization effect when combined with radiation, it must have the best radiochemical time sequence. To explore this issue, we used in vitro cell clone culture technology to observe the effect of 5-Fu+DDP combined with radiation on the human lung adenocarcinoma Anip973 cells at different time phases before, during, and after radiation for 5 hours. As a result, the DEF values of the three radiochemical combination groups were all greater than 1, and the cell viability was significantly lower than that of single-irradiation and simple drug administration. The cell killing multiple of the prodrug post-irradiation group was 4.57, which was greater than that of the intermediate irradiation group and the first irradiation. Group; DEF values were highest in the prodrug group (2.2); the average survival rate of the cells was lower in the first group than the other two groups, 0.096, 0.125, and 0.115, respectively. The results showed that the sensitizing effect of 5-Fu+DDP combined with radiation was given before radiotherapy and maintained a certain drug action time, and the best effect was obtained.
