目的 探讨 β型趋化因子MCP -1(monocyteschemotacticprotein -1,MCP -1)在充血性心力衰竭发病中的作用。 方法 用RT -PCR检测 2 5例充血性心力衰竭患者和 10例正常人外周血单个核细胞MCP -1的表达 ,并用Bio -Rad图像分析系统对其进行相对定量 ,相关分析法确定MCP -1与充血性心力衰竭各临床指标之间的关系。结果 心力衰竭患者MCP -1的表达水平与正常对照组相比显著上调 (P <0 0 1) ;源于高血压、冠心病和扩心病的心力衰竭患者MCP -1表达水平与对照组相比均有显著性差异(P <0 0 1) ;扩张型心肌病心力衰竭患者MCP -1的表达水平显著高于高血压性心力衰竭组 (P <0 0 5 )。MCP -1的表达水平与外周血白细胞计数呈正相关 (γ =0 473,P <0 0 5 ) ,与淋巴细胞计数呈正相关 (γ =0 42 5 ,P <0 0 5 ) ,而与心力衰竭患者的射血分数呈负相关 (γ =-0 64 8,P <0 0 5 )。结论 充血性心力衰竭患者外周血趋化因子MCP -1表达上调可能是心力衰竭发生的病理生理因素之一 ,其机制可能与白细胞的活化有关
Objective To investigate the role of monocyte chemotactic protein-1 (MCP-1) in the pathogenesis of congestive heart failure. Methods The expression of MCP-1 in peripheral blood mononuclear cells of 25 patients with congestive heart failure and 10 normal controls were detected by RT-PCR. Bio-Rad image analysis system was used to determine the expression of MCP-1 And the relationship between the clinical indicators of congestive heart failure. Results The expression of MCP-1 in heart failure patients was significantly up-regulated compared with the control group (P <0.01). The expression of MCP-1 in heart failure patients with hypertension, coronary heart disease and heart disease was significantly higher than that in the control group (P <0.01). The expression of MCP-1 in patients with dilated cardiomyopathy was significantly higher than that in patients with hypertensive heart failure (P <0.05). The expression of MCP-1 was positively correlated with peripheral blood leukocyte count (γ = 0 473, P 0 05) and positive correlation with lymphocyte count (γ = 0 42 5, P 0 05) The patient’s ejection fraction was negatively correlated (γ = -0 64 8, P <0 05). Conclusion Upregulation of chemokine MCP-1 in patients with congestive heart failure may be one of the pathophysiological factors of heart failure, which may be related to the activation of leukocytes
