考察 13种大孔吸附树脂对蒲黄总黄酮的吸附分离性能 ,确定大孔吸附树脂吸附分离蒲黄总黄酮的工艺条件。以总黄酮吸附量、总黄酮含量和总黄酮回收率为考察指标 ,测定总黄酮含量采用紫外 -可见分光光度法 ,考察泄露曲线采用薄层扫描法。结果表明AB - 8型树脂对蒲黄总黄酮有良好吸附分离性能 ,其吸附分离蒲黄总黄酮的工艺条件为 :蒲黄提取物上样浓度为 2 0mg/mL ,蒲黄总黄酮最大吸附量为 9 36mg/mL ,吸附流速为 2mL/min ,洗脱剂为 5 0 %乙醇 ,洗脱剂用量为 7倍量树脂体积 ,洗脱速度为 2mL/min ,树脂可重复使用 3次。AB - 8型大孔吸附树脂在所确定的工艺条件下 ,可较好地吸附分离蒲黄总黄酮。其乙醇洗脱物中蒲黄总黄酮含量达 4 5 %以上 ,总黄酮回收率达 85 %以上。
The adsorption and separation performance of 13 kinds of macroporous resin on the total flavonoids of Pueraria humilis was investigated, and the conditions for adsorption and separation of total flavonoids from Puhuang by macroporous resin were determined. Total flavonoids, total flavonoids, and total flavonoids were used as indicators for the determination of total flavonoids. UV-visible spectrophotometry was used to determine the leakage curve using thin-layer scanning. The results showed that AB-8 resin had good adsorption and separation performance for the total flavonoids of Pueraria humilis. The adsorption and separation conditions of the total flavonoids from Pueraria humilis were as follows: the concentration of Puhuang extract was 20 mg/mL, and the maximum adsorption of total flavonoids in Puhuang was obtained. It was 9 36 mg/mL, the adsorption flow rate was 2 mL/min, the eluent was 50 % ethanol, the eluent amount was 7 times the volume of the resin, the elution rate was 2 mL/min, and the resin could be reused 3 times. The AB-8 macroporous adsorption resin can be used to adsorb and separate total flavonoids from Pueraria humilis under the determined conditions. The content of total flavonoids in the ethanolic eluate was over 45%, and the recovery rate of total flavonoids was over 85%.
