目的:初步探讨远志皂苷元神经营养作用的机制。方法:体外培养新生大鼠皮质神经元,采用RT-PCR法对MAP2 mRNA及BDNF mRNA表达情况进行测定,进一步观察远志皂苷元的神经营养作用。结果:远志皂苷元低、中及高剂量均可明显提高细胞的活力,并呈一定的剂量依赖性;与溶媒对照组比较,远志皂苷元各剂量组MAP2mRNA及BDNF mRNA表达量均增加,以高剂量组(2μmol/L)最为显著(P<0.01)。结论:远志皂苷元能增加新生大鼠皮质神经元MAP2 mRNA及BDNF mRNA的表达,具有神经营养作用。
Objective: To investigate the neurotrophic mechanism of tenebrio saponins. Methods: The cortical neurons of neonatal rats were cultured in vitro. The expression of MAP2 mRNA and BDNF mRNA was determined by RT-PCR, and the neurotrophic effects of tectoridin were further observed. Results: Polygalacetin sapogenin could significantly increase the cell viability in a dose-dependent manner at low, medium and high doses. Compared with the vehicle control group, MAP2 mRNA and BDNF mRNA expression increased The dose group (2μmol / L) was the most significant (P <0.01). CONCLUSION: Polygala root sapogenin can increase the expression of MAP2 mRNA and BDNF mRNA in cortical neurons of neonatal rats and has neurotrophic effects.
