目的分析宫颈癌细胞凋亡及miRNA-34a表达与翡翠贻贝多糖的关系。方法利用翡翠贻贝多糖(100mg/L)对宫颈癌细胞进行处理,48h后,采用流式细胞术对宫颈癌细胞凋亡情况进行分析,采用实时定量PCR法对宫颈癌细胞的miRNA-34a表达进行检测。结果给药48h后,实验组宫颈癌细胞凋亡率和miRNA-34a表达相对值分别为(37.2±6.8)%、(2.5 4±0.5 2),均高于对照组的(7.7±2.4)%、(1.14±0.12),两组差异比较具有显著性(P<0.05)。结论翡翠贻贝多糖对诱导宫颈癌细胞凋亡、上调miRNA-34a表达具有重要意义。
Objective To analyze the relationship between the apoptosis of cervical cancer cells and the expression of miRNA-34a and the polysaccharide of Myrica rubra. Methods Cervical cancer cells were treated with Emerald mussel polysaccharides (100mg / L) for 48 hours. The apoptosis of cervical cancer cells was analyzed by flow cytometry. The expression of miRNA-34a in cervical cancer cells was detected by real- Test. Results The apoptotic rate and the relative expression of miRNA-34a in experimental group were (37.2 ± 6.8)% and (2.54 ± 0.52), respectively, 48.7% and 48.4% higher than those in control group (7.7 ± 2.4% , (1.14 ± 0.12) respectively. There was significant difference between the two groups (P <0.05). Conclusion Emerald mussel polysaccharide can induce cervical cancer cell apoptosis and up-regulate the expression of miRNA-34a.
