目的:研究野葛中异黄酮的纯化工艺,从而为葛根异黄酮的利用和生产提供参考。方法:通过静态吸附与解吸试验优选树脂,通过泄露曲线考察、洗脱醇浓度考察、洗脱醇用量考察等动态吸附与解吸试验优选纯化工艺。结果:葛根有效组分葛根总异黄酮富集纯化工艺为:浓度0.2g药材·mL-1的上样药液,采用AB-8树脂湿法装柱,以药材树脂用量比0.4∶1.0上柱,以8倍量50%乙醇洗脱,收集洗脱液8BV,水浴蒸干,即得葛根总异黄酮富集物。结论:葛根总异黄酮类成分的纯化工艺采用弱级性的AB-8大孔吸附树脂效果最佳。
OBJECTIVE: To study the purification technology of isoflavones in Pueraria lobata so as to provide a reference for the utilization and production of isoflavones in Pueraria lobata. Methods: The resin was selected by static adsorption and desorption test, and the purification process was optimized by dynamic adsorption and desorption tests, such as leaking curve, elution alcohol concentration, eluting alcohol content. Results: The enrichment and purification of total isoflavones of Radix puerarae puerariae was as follows: concentration of 0.2 g of drug · mL-1 sample solution, packed with AB-8 resin wet column, the ratio of drug resin to 0.4:1.0 , 8 times the amount of 50% ethanol elution, collecting eluent 8BV, water bath evaporated to obtain Pueraria total isoflavone enrichment. Conclusion: The purification technology of total isoflavones from Pueraria lobata is best by using weak AB-8 macroporous resin.
