目的观察地理位置对中华白蛉基因多态性及形态学的影响.方法选择3地采集的5组中华白蛉,用扫描电镜观察了它们的咽甲,并用5种单一的10bp引物对其基因组DNA随机扩增多态性进行了比较.结果在扫描电镜下,5组中华白蛉的咽甲形态基本一致,各组间未见明显不同.3叶咽甲的形态也无大的区别.5组白蛉的基因组DNA共扩增出138个条带,有些条带为5组白蛉所共有,大多为组间共有,个别为个体特异性条带.这样,受检的3地5组白蛉就可根据其由数种单一10bp引物扩增的基因多态性图谱区分开来.聚类分析结果表明:四川南坪的中华白蛉其亲缘关系介于山西吉县和陕西宜川的白蛉之间.结论3地采集的5组中华白蛉虽然在形态上有某些差别,但仍属同一生物种.“,”Aim To study the effect of geographical position on the genomic DNA and morphology of Phlebotomus chinensis. Method The pharyngeal armatures of 5 groups Phlefotomus chinensis from 3 different region were observed by scanning electron microscopy and their genetic DNA was ampified using 10-base random primers. Results The pharyngeal armatures were identified and the mophorlogy of the 3 leaf was alike. The 138 bands were amplified by 5 single 10-base primers, some bands were appeared in 5 groups, some in individuals, but most in more groups. After cluster analysis by computer, the 5 groups of sandflies were individed into two main groups. It is interested that the sandfly from Nanping closely related with Shanxi, and distantly from Yichuan. Conclusion Though these sandflies came from different regions, their belong to one biological species.
