目的:探讨CD4~+CD25~+调节性T细胞(CD4~+CD25~+T regs)与Foxp3在头颈部鳞状细胞癌患者外周血中的表达及意义。方法:收集2014-01-2015-12期间初诊或治疗后复发的40例头颈部鳞状细胞癌患者,均接受了手术治疗,同时纳入10例健康志愿者作为对照组。均采外周血2 ml进行流式细胞仪检测,分别计算CD4~+CD25~+/CD4~+以及CD4~+CD25~+Fxop3~+/CD4~+的比值,利用SPSS 23.0软件对结果进行统计学分析。结果:CD4~+CD25~+T regs在头颈肿瘤组织中均有高表达,与对照组比较差异有统计学意义(P<0.05)。在CD4~+CD25~+T regs阳性表达的单个细胞中,Foxp3阳性率较高,与对照组比较差异有统计学意义(P<0.01);而CD4~+CD25~+T regs在中晚期与早期头颈肿瘤中的阳性率差异有统计学意义(P<0.05)。CD4~+CD25~+T regs的表达与Foxp3的表达之间呈显著正相关(r=0.95)。结论:CD4~+CD25~+T regs与Foxp3在头颈部鳞状细胞癌患者外周血中高表达,通过对头颈部鳞状细胞癌患者免疫系统的抑制,促进了头颈部鳞状细胞癌的进展。
Objective: To investigate the expression of CD4 ~ + CD25 ~ + regulatory T cells (CD4 ~ + CD25 ~ + T regs) and Foxp3 in peripheral blood of patients with head and neck squamous cell carcinoma. Methods: Forty patients with head and neck squamous cell carcinoma who had newly diagnosed or relapsed after 2014-01-2015-12 were enrolled in the operation. Ten healthy volunteers were enrolled as the control group. The blood samples were collected from 2 ml of peripheral blood for flow cytometry. The ratios of CD4 ~ + CD25 ~ + / CD4 ~ + and CD4 ~ + CD25 ~ + Fxop3 ~ + / CD4 ~ + were calculated and the results were analyzed by SPSS 23.0 software Analysis. Results: The levels of CD4 ~ + CD25 ~ + T regs in both head and neck tumor tissues were significantly higher than those in control group (P <0.05). The positive rate of Foxp3 was higher in single cells with positive CD4 ~ + CD25 ~ + T regs expression than that in control group (P <0.01), while in CD4 ~ + CD25 ~ + T regs The positive rate of head and neck cancer was statistically significant (P <0.05). There was a significant positive correlation between the expression of CD4 ~ + CD25 ~ + Tregs and the expression of Foxp3 (r = 0.95). Conclusion: CD4 ~ + CD25 ~ + T regs and Foxp3 are highly expressed in the peripheral blood of patients with squamous cell carcinoma of the head and neck. The suppression of the immune system in patients with squamous cell carcinoma of the head and neck promotes the development of squamous cell carcinoma of the head and neck progress.
