目的 研究周期素依赖性蛋白激酶 5 (CDK5 )的mRNA及蛋白在大鼠发育过程中脑内的表达及分布。方法 采用胚胎至老年期Wistar大鼠脑片行原位杂交组织化学和免疫细胞化学染色。 结果 1 原位杂交结果显示 ,脑内CDK5mRNA在大鼠E14~P35 0整个发育过程中均有表达 ,成年后趋于稳定 ;脑内CDK5mRNA主要定位于神经元 ,阳性区域主要分布于大脑皮层、海马、丘脑、下丘脑、小脑及部分神经核团。 2 免疫组织化学结果显示 ,出生后脑内CDK5蛋白表达较强 ,胚胎及老年鼠表达较弱 ;阳性区域集中在室周区、海马、小脑及部分神经核团内 ;老年鼠仅在海马、小脑蒲肯野细胞层内表达。 结论 以上结果进一步证明 ,脑内CDK5的作用贯穿了整个神经发育的各个时期 ;老年大鼠海马内CDK5的表达下调可能与老年性学习记忆减退的发生密切相关
Objective To study the expression and distribution of CDK5 mRNA and protein in the rat brain during development. Methods In situ hybridization and immunocytochemical staining were performed in the brain of Wistar rats from embryonic to old age. Results 1 In situ hybridization showed that CDK5 mRNA was expressed throughout E14-P35 in rats and stabilized in adult rats. CDK5 mRNA mainly located in neurons in brain, mainly in cerebral cortex and hippocampus , Thalamus, hypothalamus, cerebellum and part of the nucleus. The results of immunohistochemistry showed that the expression of CDK5 protein in postnatal brain was stronger than that in embryonic and aged mice. The positive area was concentrated in the periventricular zone, the hippocampus, the cerebellum and part of the nerve nuclei. The aged mice only expressed in the hippocampus and cerebellum Ken wild cells layer expression. Conclusion The above results further prove that the role of CDK5 in the brain runs through all stages of neurodevelopment. The down-regulation of CDK5 expression in the hippocampus may be closely related to the decline of learning and memory in senile patients
