目的:观察右归饮协同骨髓干细胞(MSCs)移植对液氮冷冻股骨头坏死家兔模型骨修复的影响。方法:40只兔随机分为4组:A组(模型组)、B组(右归饮组)、C组(干细胞移植组)、D组(右归饮加干细胞移植组)。分离、培养、增殖MSCs;改良液氮冷冻法造模成功1周后,B组采用右归饮灌胃治疗,C组行髓芯减压术后MSCs 1mL(细胞密度:1×105·mL-1)移植治疗,D组采用右归饮协同MSCs移植治疗,对一般形态学、4、7周后的股骨头组织病理学和RT-PCR观察VEGF表达情况进行检测。结果:各治疗组较A组的观测指标均有明显改善,其中各治疗组空骨陷窝阳性率和VEGF阳性表达与A组组间比较差异均有统计学意义(P<0.01),且D组改善优于B、C两组(P<0.05)。结论:右归饮协同骨髓干细胞(MSCs)移植法能够显著改善冷冻坏死股骨头的血供、增强VEGF表达,促进坏死骨修复。
Objective: To observe the effects of Yougui Yin combined with bone marrow stem cells (MSCs) transplantation on repair of rabbit model bone in frozen-thawed femoral head with liquid nitrogen. Methods: Forty rabbits were randomly divided into 4 groups: group A (model group), group B (group Guowuyin), group C (group of stem cells transplantation), group D (group of Guoguxin plus stem cells). MSCs were isolated, cultured and proliferated. One week after the modified liquid nitrogen freezing method was successfully established, group B was treated with Yougui Yin intragastrically. In group C, MSCs 1 mL (cell density: 1 × 105 · mL- 1) transplantation. Group D was treated with Yougui Yin and MSCs transplantation. The morphological changes of the femoral head were observed 4 weeks and 7 weeks later. The VEGF expression was detected by RT-PCR. Results: The observation indexes in each treatment group were significantly improved compared with those in group A, and the positive rate of bone empty nest and the positive expression of VEGF in each treatment group were significantly different from those in group A (P <0.01), and D Group improved better than B, C two groups (P <0.05). Conclusion: Yougui Yin combined with bone marrow stem cells (MSCs) transplantation can significantly improve the blood supply of frozen necrotic femoral head, enhance VEGF expression and promote necrotic bone repair.
