目的探讨健脾化湿泄浊方对实验性高尿酸血症大鼠模型肾功能及黄嘌呤氧化酶(XOD)活性的影响。方法 SD大鼠60只,随机分成6组:正常对照组,模型组,健脾化湿泄浊方高、中、低剂量组和阳性对照(别嘌醇)组,每组10只。建立腺嘌呤+氧嗪酸钾致高尿酸血症大鼠模型,同时连续给药30 d。在给药前及给药第30天取血,分离血清测定尿酸(UA)、尿素氮(BUN)、肌酐(Cr)和XOD活性,并取动物肾脏作HE染色。结果与模型组比较,健脾化湿泄浊方高、中剂量组在给药30 d后可显著降低高尿酸模型大鼠血UA水平、血清XOD活性(P<0.01);血清BUN、Cr的检测结果和肾组织病理学检查结果显示,健脾化湿泄浊方高、中剂量组对高尿酸血症大鼠肾功能有一定的保护作用。结论健脾化湿泄浊方具有良好的降尿酸作用,对肾功能有一定的保护作用,其降尿酸作用可能与其抑制XOD活性有关。
Objective To investigate the effects of Jianpi Huashi Xiezhuo prescription on renal function and xanthine oxidase (XOD) activity in experimental hyperuricemic rats. Methods Sixty SD rats were randomly divided into 6 groups: normal control group, model group, Jianpi Huashi Xiezhuo Fang high, medium and low dose group and positive control (allopurinol group), 10 rats in each group. To establish a rat model of hyperuricemia induced by adenine and oteracil potassium, and administrated for 30 days continuously. Before administration and on the 30th day of administration, the blood was collected and the serum was measured for UA, BUN, Cr and XOD activity. The kidneys were taken for HE staining. Results Compared with the model group, the serum UA level and the serum XOD activity in serum of rats with hyperuricemia model (P <0.01) were significantly decreased after 30 days of administration, Test results and renal histopathological examination showed that the spleen Huashi Xiexie Fang high and medium dose group of renal function in rats with hyperuricemia have a protective effect. Conclusions Jianpi Huashi Xiezhuo Recipe has a good effect of lowering uric acid and has a protective effect on renal function. Its uricosuric effect may be related to its inhibition of XOD activity.
