目的观察葛根素(Pue)对局灶脑缺血大鼠脑源性神经营养因子(BDNF)表达的影响,进一步探讨葛根素的神经保护作用。方法制作大鼠大脑中动脉闭塞(MCAO)模型,30只SD雄性大鼠被随机分为假手术组、缺血组、Pue治疗组。应用TTC染色观察梗死体积,TUNEL染色检测凋亡细胞数,免疫组化方法观察Pue治疗组及脑缺血组BDNF阳性细胞数,并进行图像分析。结果与假手术组相比,缺血组及Pue治疗组BDNF阳性细胞均显著增加。且Pue治疗组阳性细胞数又显著高于缺血对照组(P<0.05)。结论Pue的神经保护作用可能与其能提高内源性BDNF的表达水平有关。
Objective To observe the effect of puerarin on the expression of brain-derived neurotrophic factor (BDNF) in focal cerebral ischemia rats and further explore the neuroprotective effect of puerarin. Methods The middle cerebral artery occlusion (MCAO) model was established in rats. Thirty SD male rats were randomly divided into sham operation group, ischemia group and Pue treatment group. The infarct volume was observed by TTC staining, the number of apoptotic cells was detected by TUNEL staining, and the number of BDNF positive cells in Pue treatment group and cerebral ischemic group was observed by immunohistochemistry method, and image analysis was performed. Results Compared with the sham group, BDNF-positive cells were significantly increased in the ischemic group and the Pue treated group. The number of positive cells in the Pue treatment group was significantly higher than that in the ischemic control group (P<0.05). Conclusion The neuroprotective effect of Pue may be related to its ability to increase the expression of endogenous BDNF.
