目的:考察丹黄凝胶提取物及其指标成分大黄酚和小檗碱在外用制剂常用溶媒中的溶解性,同时测定提取物中大黄酚和小檗碱的油水分配系数。方法:运用沉淀法、指标成分溶解量法测定提取物及其主要活性成分大黄酚和小檗碱的溶解性,采用摇瓶法测定提取物中大黄酚和小檗碱的油水分配系数。结果:丹黄凝胶提取物及大黄酚和小檗碱在丙二醇中溶解性最好,溶解度分别为8.25,60.17,353.01 mg.L-1;提取物中大黄酚和小檗碱的油水分配系数受pH影响较大。结论:丙二醇和乙醇能较好地溶解提取物及其活性成分大黄酚和小檗碱;适宜pH可使大黄酚和小檗碱具有一定的脂溶性和水溶性,有利于其透皮吸收。
OBJECTIVE: To investigate the solubility of extract of Dan Huang gel and its index components chrysophanol and berberine in common solvents for external application, and to determine the oil-water partition coefficients of chrysophanol and berberine in the extract. Methods: Solubility of chrysophanol and berberine were determined by precipitation method and index component dissolution method. The oil-water partition coefficients of chrysophanol and berberine were determined by shake flask method. Results: The extract of chrysanthemum morifolium and chrysophanol and berberine had the best solubility in propylene glycol with the solubilities of 8.25, 60.17 and 353.01 mg.L-1, respectively. The oil-water partition coefficient of chrysophanol and berberine Affected by pH. CONCLUSION: Propylene glycol and ethanol can dissolve the extract and its active components, chrysophanol and berberine. Appropriate pH can make chrysophanol and berberine have some liposolubility and water solubility, which is good for transdermal absorption.
