目的探讨重楼皂苷Ⅰ对IL-17刺激Ha Ca T细胞后VEGF和IL-23表达的影响。方法 CCK-8检测各组Ha Ca T细胞活力,采用ELISA方法和荧光实时定量PCR(RT-PCR)方法检测上清中VEGF,IL-23的含量及VEGF mRNA,IL-23 mRNA的表达。结果培养24h,CCK-8检测重楼皂苷Ⅰ+IL-17组及IL-17组Ha Ca T细胞活力无明显差异(P>0.05)。与对照组相比,IL-17(80ng/m L)刺激组上清中VEGF,IL-23的含量及Ha Ca T细胞VEGF mRNA,IL-23 mRNA表达均高于对照组(P<0.05)。重楼皂苷Ⅰ(1.5μg/m L,3μg/m L)可抑制IL-17刺激Ha Ca T细胞后上清VEGF,IL-23的含量及VEGF mRNA,IL-23mRNA的表达(P<0.05)。结论 IL-17可促进Ha Ca T细胞分泌VEGF,IL-23和表达VEGF mRNA,IL-23 mRNA,而重楼皂苷Ⅰ对其具有抑制作用,该结果为重楼皂苷Ⅰ抑制IL-17介导的皮肤免疫性炎症提供了一定的依据。
Objective To investigate the effect of Pollen saponin Ⅰ on the expression of VEGF and IL-23 after stimulating HaCa T cells with IL-17. Methods The activity of HaCa T cells in each group was detected by CCK-8. The contents of VEGF and IL-23 and the expressions of VEGF mRNA and IL-23 mRNA in supernatants were detected by ELISA and real-time quantitative PCR (RT-PCR) Results After incubated with CCK-8 for 24 h, there was no significant difference in the activity of HaCa T cells between RSPS + IL-17 and IL-17 (P> 0.05). Compared with the control group, the levels of VEGF and IL-23 in the supernatant of IL-17 (80ng / m L) stimulation group and the expressions of VEGF mRNA and IL-23 mRNA in HaCa T cells were higher than those in the control group (P <0.05) . The contents of VEGF and IL-23 and the expressions of VEGF mRNA and IL-23 mRNA in supernatant of HaCa T cells stimulated by IL-17 were significantly inhibited by 1.5 g / ml and 3 μg / ml LPS, respectively (P <0.05) . Conclusion IL-17 can promote the secretion of VEGF, IL-23 and express VEGF mRNA and IL-23 mRNA in HaCa T cells, and the inhibitory effect of polysaccharide Saponins Ⅰ on them. The skin immune inflammation provides a basis.
