目的:了解p53基因突变与埃伯斯坦-巴尔病毒(EBV)相关性鼻咽部T细胞淋巴瘤(NPT)的关系。方法:应用免疫组织化学方法(SABC),原位杂交(ISH),PCR-SSCP对26例确诊为鼻咽部T细胞淋巴瘤的病例进行p53蛋白及基因的检测。结果:EBV编码的小RNA探针(EBER)17/26例阳性(654%);p53蛋白13/26例阳性(50%);对其中12例进行p53外显子5,7,8的PCR-SSCP检测,发现p53突变7/12例(583%)阳性,EBV阳性组5/8例阳性(625%),阴性组2/4例(50%)阳性,外显子5突变4/12例(333%)阳性,外显子7突变1/12例(83%)阳性,外显子8突变3/12例(25%)阳性,其中1例同时发生5,8两个外显子突变;EBER/p53蛋白表达同时发生有11例,不发生7例,EBER+/p53-6例,EBER-/p53+2例;EBER表达(ISH)与p53基因突变(PCR-SSCP)同时发生5例,不发生2例,EBER+/p53-(PCR-SSCP)3例,EBER-/p53+(PCR-SSCP)2例;以p53蛋白检测为标准,p53基因突变(PCR-SSCP?
Objective: To investigate the relationship between p53 gene mutation and EBV-associated nasopharyngeal T-cell lymphoma (NPT). Methods: Twenty-six cases of nasopharyngeal T-cell lymphoma were detected by immunohistochemistry (SABC), in situ hybridization (ISH) and PCR-SSCP. RESULTS: 17/26 EBV-encoded small RNA probes (EBRs) were positive (65.4%) and 13/26 (50%) of p53 proteins were positive.
