目的探讨内嗅皮层注射Orexin-A对Sprague-dawley(SD)大鼠睡眠-觉醒周期以及对空间记忆的影响。方法选用健康成年雄性SD大鼠45只,体质量250~300 g,采用同步记录大鼠脑电和肌电活动,观察Orexin-A及Orexin1受体阻断剂SB-334867内侧内嗅皮层微注射后大鼠睡眠-觉醒周期变化。采用T-迷宫交替选择次数,观察SB-334867内侧内嗅皮层微注射后大鼠T-迷宫选择正确率的变化。结果以人工脑脊液(ACSF)、二甲亚砜(DMSO)自身对照相比,分别给予大鼠内侧内嗅皮层双侧微注射Orexin A及SB-334867 0.5μl后3 h(11:00-12:00、12:00-13:00、13:00-14:00)觉醒周期时间无影响(P>0.05),给药后24 h觉醒周期时间也无显著差异(P>0.05)。微量注射药物后,Orexin-A(0.1 nmol/0.5μl)及SB-733467(6 mmol/0.5μl)在11:00-14:00 3 h内对觉醒时间、NREM时间及REM时间均无影响(P>0.05),即对3 h内(11:00-14:00)大鼠总睡眠觉醒时间无显著性差异(P>0.05)。与DMSO组比较大鼠内侧内嗅皮层双侧微注射SB-334867 0.5μl后,T-迷宫交替选择正确次数明显减少,正确率下降,有显著性差异(P<0.001)。结论内侧内嗅皮层Orexin系统的支配可能参与了空间记忆,而不参与睡眠-觉醒周期调节。
Objective To investigate the effect of Orexin-A injection in entorhinal cortex on sleep-wake cycle and spatial memory in Sprague-dawley (SD) rats. Methods Forty-five healthy adult male Sprague-Dawley rats weighing 250-300 g were used to record the changes of electroencephalogram (EEG) and electromyography (EEG) activities of the rats. The inner entorhinal cortex of Orexin-A and Orexin1 receptor antagonist SB-334867 were microinjected After the rat sleep - wake cycle changes. T-maze was used to select the number of times alternately to observe the changes of the correct selection rate of T-maze in rats after microinjection of SB-334867 into the medial entorhinal cortex. Results Compared with ACSF and DMSO self-control group, Orexin A and SB-334867 bilateral microinjection of 0.5μl into the medial entorhinal cortex were given at 3h (11: 00-12: (P> 0.05). There was no significant difference in awakening cycle time at 24 h after administration (P> 0.05). After microinjection, Orexin-A (0.1 nmol / 0.5 μl) and SB-733467 (6 mmol / 0.5 μl) had no effect on awake time, NREM time and REM time within 11: 00-14: 00 3 h P> 0.05), that is within 3 h (11: 00-14: 00), there was no significant difference in total sleep arousal time (P> 0.05). Compared with the DMSO group, the number of double-sided SB-334867 microinjection in the medial entorhinal cortex of rats was significantly reduced (P <0.001). Conclusion The dominant inner entorhinal cortex Orexin system may be involved in spatial memory, not involved in sleep-wake cycle regulation.
