目的:探讨Duffy抗原基因rs863002和rs3027012位点的单核苷酸多态性(SNP)与结直肠癌易感性的关系。方法:收集100例结直肠癌患者和100例健康对照者的外周血样本,提取全血DNA;选择Duffy抗原基因2个具有代表性的标签SNP位点(rs863002和rs3027012),使用TaqMan-MGB探针Real-time PCR技术对样本进行基因分型。结果:rs863002位点的CC基因型频率在结直肠癌组和对照组分别为90.0%和88.0%,CT基因型频率分别为10.0%和12.0%,两组间差异无统计学意义(P>0.05)。rs863002位点的C等位基因频率在结直肠癌组和对照组中分别为95.0%和94.0%,T等位基因频率分别为5.0%和6.0%,两组间差异无统计学意义(P>0.05)。rs3027012位点的CC基因型频率在结直肠癌组和对照组分别为97.0%和98.0%,CT基因型频率分别为3.0%和2.0%,两组间差异无统计学意义(P>0.05)。rs3027012位点C等位基因频率在结直肠癌组和对照组中分别为98.5%和99.0%,T等位基因频率分别为1.5%和1.0%,两组间差异无统计学意义(P>0.05)。结论:尚未发现Duffy抗原基因rs863002和rs3027012位点SNP与结直肠癌易感性存在关联。 Objective: To investigate the relationship between single nucleotide polymorphisms (SNP) of rs863002 and rs3027012 in Duffy antigen gene and susceptibility to colorectal cancer. Methods: Peripheral blood samples from 100 patients with colorectal cancer and 100 healthy controls were collected and the whole blood DNA was extracted. Two representative SNPs (Ducky’s antigenic SNPs) (rs863002 and rs3027012) were selected, and TaqMan-MGB probe Real-time PCR techniques were used to genotype samples. Results: The frequencies of CC genotypes in rs863002 locus were 90.0% and 88.0% in colorectal cancer patients and controls, respectively. The frequency of CT genotypes was 10.0% and 12.0% respectively, with no significant difference between the two groups (P> 0.05 ). The frequency of allele C at rs863002 was 95.0% and 94.0% in colorectal cancer group and control group respectively, and the frequencies of T allele were 5.0% and 6.0% respectively. There was no significant difference between the two groups (P> 0.05). The frequency of CC genotypes in rs3027012 was 97.0% and 98.0% in colorectal cancer patients and controls, respectively. The frequencies of CT genotypes were 3.0% and 2.0% respectively. There was no significant difference between the two groups (P> 0.05). The frequency of allele C at rs3027012 was 98.5% and 99.0% in colorectal cancer group and control group, respectively. The frequencies of T allele were 1.5% and 1.0% respectively, with no significant difference between the two groups (P> 0.05 ). Conclusion: There is no association between Duffy antigen gene rs863002 and rs3027012 SNP and colorectal cancer susceptibility.
