目的观察羟喜树碱纳米制剂对原位肝癌小鼠的抗肿瘤作用。方法造模小鼠30只随机分成3组,每组10只,羟喜树碱纳米组予羟喜树碱纳米制剂5 mg.kg-1;羟喜树碱粉针剂组予羟喜树碱冻干粉针剂5 mg.kg-1;对照组予等量生理盐水,每周d 1、4尾静脉给药,共2 wk。观察原位肝癌的生长状态及组织病理学改变、计算抑瘤率、MTT法检测细胞增殖抑制率及流式细胞仪检测细胞凋亡率。结果与对照组比较,羟喜树碱粉针剂组及纳米组均可抑制小鼠原位肝癌的生长,其羟喜树碱纳米组抑瘤率为48.76%,高于粉针剂组(38.51%);羟喜树碱纳米组对肝癌细胞及正常肝细胞的增殖抑制率分别为82.63%、27.10%,对瘤组织细胞凋亡率为(49.72±7.51)%。结论羟喜树碱纳米制剂可抑制小鼠原位肝癌生长。
Objective To observe the anti-tumor effect of hydroxycamptothecin nano-preparation on orthotopic liver cancer mice. Methods Thirty mice were randomly divided into three groups (n = 10 in each group), and received hydroxycamptothecin (NM) 5 mg.kg-1. Hydroxycamptothecin Dry powder injection 5 mg.kg-1; control group was given the same amount of saline, weekly d4 d vein for 2 wk. The growth and histopathological changes of HCC were observed. The tumor inhibition rate was calculated. The cell proliferation inhibition rate was detected by MTT assay and the apoptosis rate was detected by flow cytometry. Results Compared with the control group, both hydroxycamptothecin powder injection group and nano-group could inhibit the growth of orthotopic liver cancer in mice. The inhibition rate of hydroxycamptothecin nano-group was 48.76%, higher than that of the injection group (38.51%), The inhibitory rates of hydroxycamptothecin nano-particles on proliferation of hepatocellular carcinoma cells and normal hepatocytes were 82.63% and 27.10%, respectively. The apoptosis rates of hepatocellular carcinoma cells were (49.72 ± 7.51)%. Conclusion Hydroxy camptothecin nano-preparation can inhibit the growth of orthotopic liver cancer in mice.
