研究造血微环境(HME)的生理功能及其在各种血液病中的病理改变,对认识血液病的发病机理,提高临床诊治水平有着重要的意义。本研究利用骨髓基质细胞(F—CFU)体外培养、造血干细胞培养、细胞染色体分析、电镜观察和羟基脲自杀试验等方法,结合临床对HME的某些生理功能及其在一些血液病发病机理中的作用作了广泛的探讨。1、首先在国内建立了人骨髓F—CFU培养的方法,探索了细胞浓度和小牛血清等条件对F—CFU生长的影响。证明用低浓度细胞小平皿短期液体培养法是定理骨髓的F—CFU的方便、稳定、可靠且经济的方法,较适于再障等骨髓增生低下病人的测定;长期液体培养是做F—CFU细胞染色体分析的可靠
To study the physiological function of hematopoietic microenvironment (HME) and its pathological changes in various blood diseases is of great significance for understanding the pathogenesis of hematological diseases and improving the level of clinical diagnosis and treatment.
