在急性毒性试验的基础上,选用SPF级C57BL/6J小鼠50只,随机分为5组,每组10只,雌雄各半。胡桃楸种仁壳黄酮组灌胃量分别为2000、4000、8000 mg/kg,2次给药,另设阴性对照组和环磷酰胺阳性对照组。2次给药后6 h取股骨骨髓做涂片、Gimsa染色,油镜下计数微核细胞。胡桃楸种仁壳黄酮3个剂量组与阴性对照组比较,MPCE比率未显著增加(p>0.05),并极显著(p<0.01)低于阳性对照组。表明胡桃楸种仁壳黄酮未引起小鼠骨髓细胞中嗜多染红细胞微核的显著增加,无体内致突变性和遗传学毒性。
On the basis of acute toxicity test, 50 mice of SPF grade C57BL / 6J were randomly divided into 5 groups with 10 mice in each group. Juglans septemica flavone group gavage were 2000,4000,8000 mg / kg, 2 doses, another set of negative control group and cyclophosphamide positive control group. Six hours after the second administration, the bone marrow of the femur was smeared and stained with Gimsa, and the micronucleus cells were counted under an oil microscope. Compared with the negative control group, the MPCE ratio did not significantly increase (P> 0.05) in the three dosage groups of Juglans mandshurica flavonoids and was significantly lower (p <0.01) than the positive control group. These results indicated that the seed-hull flavonoids of Juglans mandshurica did not cause a significant increase in the micronuclei of mouse polychromatic erythrocytes in mouse bone marrow cells, and had no in vivo mutagenicity and genetic toxicity.
