目的:研究苦参碱对唾液腺腺样囊性癌ACC-M细胞黏附侵袭能力的影响并初步探讨其机制。方法:体外培养ACC-M细胞系,MTT法测定药物对细胞增殖的抑制作用,应用黏附试验和Matrigel侵袭实验,研究苦参碱注射液及其不同浓度对ACC-M细胞黏附及侵袭能力的影响,采用SPSS11.0软件包对数据进行t检验。结果:苦参碱注射液浓度在0.25、0.50、0.75、1.00 mg/mL时,对ACC-M细胞均有一定增殖抑制作用,且呈明显的量效和时效关系。不同浓度苦参碱注射液处理ACC-M细胞24h后,其体外黏附和侵袭能力呈剂量依赖性下降。经苦参碱(0.75 mg/mL)作用后,ACC-M细胞的E钙黏素(E-cad蛋白)表达明显增强,与对照组有显著差异。结论:一定浓度的苦参碱可以抑制唾液腺腺样囊性癌细胞生长,降低细胞黏附和侵袭能力,其机制可能与上调E-cad蛋白表达有关。
Objective: To investigate the effect of matrine on the adhesion and invasion of ACC-M cells in salivary adenoid cystic carcinoma and to explore its mechanism. Methods: ACC-M cell lines were cultured in vitro. MTT assay was used to determine the inhibitory effect of drugs on cell proliferation. Adhesion test and Matrigel invasion assay were used to investigate the effects of matrine injection and its different concentrations on the adhesion and invasion ability of ACC-M cells , Using SPSS11.0 software package for t test data. Results: Matrine injection at a concentration of 0.25,0.50,0.75,1.00 mg / mL ACC-M cells have a certain inhibition of proliferation, and showed a significant dose-effect and time-dependent relationship. Treatment of ACC-M cells with matrine at different concentrations for 24 h resulted in a dose-dependent decrease in the ability of adhesion and invasion in vitro. After treated with matrine (0.75 mg / mL), the expression of E-cadherin (E-cad) in ACC-M cells was significantly increased, which was significantly different from the control group. Conclusion: Matrine at a certain concentration can inhibit the growth of salivary adenoid cystic carcinoma cells and decrease the cell adhesion and invasion ability. The mechanism may be related to the up-regulation of E-cad protein expression.
