目的 探讨HSV tk GCV系统联合rhTNF α抗脑胶质瘤的有效性。方法 利用重组HSV tk逆转录病毒载体转染大鼠C6细胞得到HSV tk阳性细胞 ,C6 GCV、C6+tk GCV、C6+rhTNF α和C6+tk GCV +rhTNF α4组在体外分别给予GCV或 和rhTNF α处理 ,用MTT法测量细胞存活率 ,比较各组肿瘤细胞存活率。卡方检验作显著性检测。结果 未加任何抗癌因子处理的C6细胞组肿瘤细胞生长旺盛 ,细胞形态无明显改变 ;各处理组的肿瘤细胞 2~ 3天后开始逐渐出现细胞突触消失 ,细胞变圆 ,胞体皱缩 ,核仁固缩、裂解、甚至消失 ,继而逐渐死亡。联合应用HSV tk GCV +rhTN α组的肿瘤细胞存活率比单纯应用HSV tk GCV组或rhTNF α组明显低 (P <0 0 5)。结论 :联合应用HSV tk GCV系统和rhTNF α的杀癌细胞效果优于单一使用HSV tk GCV系统或者rhTNF α。
Objective To investigate the effectiveness of HSV tk GCV combined with rhTNF α against glioma. Methods HSV tk positive cells were obtained by transfecting rat C6 cells with recombinant HSV tk retroviral vector. GCV or rhTNF were respectively given in vitro in groups of C6 GCV, C6 + tk GCV, C6 + rhTNF α and C6 + tk GCV + rhTNF α4, α, the cell survival rate was measured by MTT method, and the survival rate of tumor cells in each group was compared. Chi-square test for significant detection. Results The tumor cells of C6 cells treated with no anti-cancer factor grew vigorously with no significant changes in cell morphology. After 2 to 3 days, tumor cells in each treatment group began to gradually disappear synapse, became round, cell body was shrunk, Ren shrinks, lysis, or even disappear, and then gradually die. The survival rate of tumor cells in combination with HSV tk GCV + rhTN α group was significantly lower than that in HSV tk GCV group or rhTNF α group alone (P <0.05). Conclusions: Combination of HSV tk GCV system and rhTNF α in killing cancer cells is superior to single HSV tk GCV system or rhTNF α.
